Based on data from animal studies, oral spironolactone may interfere with the development of primary sex characteristics in male fetuses.
Side Effects And Risks

Can you drink alcohol while taking spironolactone? It is best to limit alcohol intake while taking spironolactone, as alcohol can increase the side effects, including dizziness.
Spironolactone is a potassium-sparing diuretic. That means even though it flushes out extra water, it wont lower potassium levels.
If youre not avoiding potassium-rich foods , you could enter hyperkalemia, a state of too much potassium.
Hyperkalemia, or too much potassium, can lead to:

All participants will receive the assigned study treatment until unacceptable toxicity, death, loss to follow up, withdrawal of consent or termination of the study by the Sponsor, in accordance with local standards of care. Unacceptable toxicity and death will be considered as failure for the primary criteria analysis. Loss to follow up, missing data for the primary criteria and early termination of the study by the Sponsor will be also considered as failures by the ITTm and not replaced.
Patients who did not fulfil the inclusion criteria after randomization and those who never began their treatment will be replaced. For the clinical secondary criteria analysis, patients with missing data will be withdrawn. Missing data in cost-effectiveness analysis will be handled according to a multiple imputation method in the ITTm sample. If a patient is released from the study preventing the continuation of the study), her data will not be collected, except for safety data .
Procedure For Unblinding If Needed

The investigator should inform the Sponsor if unblinding is deemed to be necessary due to an emergency. Unblinding will be performed via the eCRF thanks to a detailed procedure that will be given to each site. Each decoding will be tracked and classified in the investigators management file and in the trial management file.

How To Use Spironolactone
Spironolactone is taken orally via pill or liquid medication. The most common dosage is between 50mg and 100mg daily, though some individuals may require up to 200mg to see changes in their skin. While it can be taken on an empty stomach, taking it with food can help prevent an upset stomach. Additionally, it is recommended that you take it earlier in the day to avoid having to get up during the night to pee.
So Which Form Is More Effective
Depending on the dose, oral spironolactone is typically more effective than topical spironolactone. In sum:

Topical spironolactone works best when paired with other treatments. It can add a powerful punch to other acne-fighting formulas, but it might not be as effective by itself. Its also better suited for individuals who arent a good candidate for oral spironolactone and those who want to avoid the potential risks of oral spironolactone.

Spironolactone Might Not Work For You
If you’ve been taking the pill for over three months and your hormonal acne situation isn’t any better, it might be time to try a new dose or another treatment option. “When using it to treat acne, spironolactone is only used in women and tends to work best for women with hormonal acne, menstrual flares, adult-onset acne, or conditions with hormone abnormalities and acne ,” explains Dr. Sejal Shah, cosmetic dermatologist and founder of Smarter Skin Dermatology in New York City “It is also often used in women whose acne is resistant to conventional therapies.”
“At a high enough dose, it has some effect on all women,” adds Dr. Zeichner. “The issue is that at higher doses it may be associated with side effects like breast tenderness or irregular periods, which get in the way with using it.”

Spironolactone For Cystic Acne: The Result Is Not Instant

You will start seeing the effect of spironolactone on cystic acne, but the result is not always instant. Yes, the product drastically reduces hormone, but the result will rarely be instant. So, you should bear this in mind.

Spironolactone can regulate your hormone levels in order to treat hormonal acne, but changing your hormones should not be your first defense against acne.
So how does Spironolactone work for acne? It suppresses the uptake of the hormones generally associated with acne. But altering your hormone levels can come with various side effects that can easily be avoided by trying a holistic skin care treatment that treats the excess oil production that causes hormonal acne.
While those experiencing hormonal acne may find some relief from this hormonal treatment or from oral contraceptives, a well-formulated acne-fighting skincare regimen is likely to be more effective without the cost and side effects that can accompany a new prescription.
Spironolactone For Acne Side Effects

Potential side effects of spironolactone include:
- Breast tenderness
- Fatigue
- Increased urination
Dr. Zeichner says many of these side effects are more likely when the medication is given at higher doses. Its worth noting that there is a black box warning on the drug, but it may not be a cause for genuine concern if you are an otherwise healthy female.
In one study in rats spironolactone caused solid organ tumors, however it has not been shown to be a problem in humans, advises Dr. Zeichner. The amount of the drug given to rats in the study was significantly higher than the one prescribed to women.
Spironolactone does not typically cause depression or mood changes. If you do notice changes in mood while taking it, let your healthcare provider know. Sometimes, spironolactone is prescribed to promote hair growth in women because its an antiandrogenthat means it blocks the male hormones that may lead to hair loss in women.
Because the drug decreases fluid retention, you may lose a small amount of weight while taking it. There isnt any evidence that spironolactone works as a weight loss medication, but it may help with premenstrual bloating and is often prescribed to treat symptoms of PMS and premenstrual dysphoric disorder .

Oral Spironolactone Is Moderately Effective In Treating Acne
Ten studies performed to date indicate that oral spironolactone is at least moderately effective in treating acne. While the studies used different methods and looked at different numbers of patients, the results of each study show at least moderate improvement in acne in the majority of patients.4-13 It should be noted that although two of the studies included male patients, doctors almost always prescribe spironolactone only to women.

A 1984 study in the British Journal of Dermatology investigated the efficacy of spironolactone in both men and women with severe acne. Although the researchers examined men, doctors almost always prescribe spironolactone only to women. They randomly assigned 36 participants to receive one of five treatments.

5. A placebo
Researchers in a 1986 study in the same journal gave 21 women 200mg of spironolactone daily for three months. The researchers randomly assigned the women to a treatment group or to a placebo group. This study found that the group taking spironolactone experienced a 66% improvement in the number of acne lesions after three months. In contrast, the women in the placebo group did not experience improvement in their acne. Seventy-three percent of the women who did not take a COC also experienced menstrual side effects, but all of the women persisted. Two women dropped out of the study because of nausea.5
Aldactone For High Blood Pressure
Aldactone is FDA-approved to treat hypertension in adults. For this purpose, Aldactone should be used with other blood pressure drugs.
A decrease in blood pressure can help lower the risk of cardiovascular problems, such as heart attack and stroke. In some cases, these problems can be fatal.
High blood pressure explained
As blood flows through arteries, it exerts a force on the artery walls. This is called blood pressure. When the force is higher than usual, its considered high blood pressure.
Many people with high blood pressure dont have any symptoms of the condition. In fact, its possible for you to have high blood pressure for many years before you notice any symptoms, if you ever have symptoms at all.
Symptoms of high blood pressure can include:
- changes in vision
- shortness of breath
If high blood pressure isnt treated, it can lead to serious complications, including damage to your arteries and heart. In rare cases, these complications can be fatal. High blood pressure can also lead to decreased blood supply to your brain, which could result in brain damage.
Effectiveness for high blood pressure
According to clinical guidelines from the American College of Cardiology and American Heart Association, spironolactone may be used with other medications to help lower blood pressure.

What Are The Side Effects Of Oral Spironolactone
As with all medications, spironolactone can cause side effects, particularly at higher doses. Most people tolerate low doses of spironolactone well, even for long periods of time. Side effects that can occur at dosages higher than 100mg per day include:
- Menstrual irregularities, such as increased or decreased menstrual bleeding or breakthrough bleeding, which is bleeding that occurs between periods. Adding a COC to the treatment reduces these side effects.
- Tenderness or enlargement of the breasts in both males and females. Adding a COC to the treatment can reduce this side effect.
- Orthostatic hypotension, which is a sudden drop in blood pressure when standing up that can result in dizziness or fainting.
- Reduced libido .
- Excessively high potassium level in the blood. This is a dangerous side effect that can cause life-threatening heart problems. However, it is not common in healthy young people. Because of this side effect, people with kidney problems should not use spironolactone. In addition, spironolactone cannot be used with certain medications, including diuretics and ACE inhibitors, which often are used for high blood pressure.3
Side effects that do not seem to be related to dose include:
- Tiredness
